The nature and function of alamethicin.

چکیده
The tubulin-rich fraction was also shown to contain a protein subunit, of mol.wt. 46000f 1500, identified as actin by co-electrophoresis with skeletal-muscle actin. Although actin can be removed from brain tubulin preparations by repeated cycles of depolymerization/polymerization, its total removal from the platelet tubulin-rich fraction is most difEcult. In addition to tubulin and actin, three higher-molecular-weight components are apparent (Fig. l), some of whichmay represent the dynein-like proteinsrecently observed in brain tubulin preparations obtained by polymerization (Gaskin e ta / . , 1974; Burns & Pollard, 1974).

Electronic control of H+ current in a bioprotonic device with Gramicidin A and Alamethicin
In biological systems, intercellular communication is mediated by membrane proteins and ion channels that regulate traffic of ions and small molecules across cell membranes. A bioelectronic device with ion channels that control ionic flow across a supported lipid bilayer (SLB) should therefore be ideal for interfacing with biological systems. Here, we demonstrate a biotic-abiotic bioprotonic de...

The effects of alamethicin on the membrane barrier function of rabbit erythrocytes, human platelets and sarcoplasmic reticulum vesicles, as well as on that of brain microsomes and liver mitochondria of the rat were compared. An upset of the barrier function was observed for plasma membranes of brain microsomes as well as for erythrocyte and platelet membranes at alamethicin concentrations rangi...
متن کاملLipid chain-length dependence for incorporation of alamethicin in membranes: electron paramagnetic resonance studies on TOAC-spin labeled analogs.
Alamethicin is a 19-residue hydrophobic peptide, which is extended by a C-terminal phenylalaninol but lacks residues that might anchor the ends of the peptide at the lipid-water interface. Voltage-dependent ion channels formed by alamethicin depend strongly in their characteristics on chain length of the host lipid membranes. EPR spectroscopy is used to investigate the dependence on lipid chain...
متن کاملLipid-alamethicin interactions influence alamethicin orientation.
Whereas the barrel-stave configuration is accepted by most investigators as a good description of the conducting state of alamethicin, there are conflicting interpretations on its nonconducting state; in the absence of an applied field, some found alamethicin molecules on the membrane surface, but others found them incorporated in the hydrophobic core of the membrane. This problem is resolved b...
متن کاملArtificial membrane excitability revisited and implications for the gating of voltage-dependent ion channels.
Excitability phenomena in planar lipid bilayers doped with alamethicin and protamines have been first described by Mueller and Rudin (Nature 217, 713-719, 1968). These properties are reinvestigated here with virtually solvent-free bilayers made of synthetic phospholipids doped with alamethicin charged component (Glu18) and protamine or other synthetic basic polypeptides. After retrieving the na...
